A series of oligosaccharides and structural characterization in atractylodes
Abstract:As a traditional Chinese medicine, Atractylodes lancea has a wide range of pharmacological effects and has a good application prospect in clinical practice. However, the chemical composition of Atractylodes is very complicated. At present, research topics for fat-soluble substances in Atractylodes macrocephala are in the majority, and there are few issues that aim at the water-soluble components contained in Atractylodes lancea. However, the clinical application of Atractylodes Rhizome is mostly decoction. Modern pharmacological results also confirm that Atractylodes aqueous extract has good The pharmacological activity. This experiment focused on the in-depth study of the oligosaccharide contained in the aqueous extract of Atractylodes. The oligosaccharides were extracted using the more common ethanol reflux method to optimize the extraction conditions and improve the extraction efficiency. At the same time, the oligosaccharides extracted from the protein purification system (AKTA) and the semi-preparative solution were separated and purified, and detected by high performance liquid chromatography (HPLC). Structural characterization of oligosaccharide extracts was then performed using acid hydrolysis coupled with liquid phase analysis and methylation analysis. The experimental results show that ethanol refluxing method can effectively extract oligosaccharides from Atractylodes Rhizome, and AKTA and semi-preparation liquids have good separation and purification effects relative to oligosaccharides. In addition, through the monosaccharide composition experiment, it was found that the atractylotic oligosaccharide was a chrysanthemum-type oligosaccharide, and the glycosidic bond type was identified by a methylation experiment.
