互花米草总黄酮的提取工艺研究毕业论文

摘 要

互花米草是一种分布较广,且药用功效丰富的植物。本文从互花米草的潜在药用价值切入,以互花米草的总黄酮化合物作为主要研究成分,从而制备互花米草总黄酮,并且对提取工艺条件进行优化,为了能进一步开发利用互花米草,本文对制备的互花米草总黄酮进行了质量评价。

采用乙醇回流提取法提取互花米草中黄酮类成分,配制芦丁标准溶液,以AlCl3和CH3COONa 作显色剂,C2H6O作溶剂,在紫外可见分光光度计上测定最大吸收波长,绘制芦丁标准曲线。优化提取工艺时,乙醇浓度、提取温度、料液比以及提取时间均由单因素法实现筛选,从而制备出符合要求的互花米草总黄酮,并对其进行质量评价。

在420nm波长的紫外可见光的检测下,芦丁标准曲线方程为Y=24.52X 0.033,其中R2=0.994。由单因素实验确定了提取时的最优乙醇浓度为70%,最优提取温度为60℃,最优料液比为1:30,最优提取时间为90min。通过对大孔树脂类型、静态吸附以及解吸附条件的筛选,确定采用AB-8型大孔树脂,吸附时间8h,解吸时间6h,解吸液体积分数80%,解吸液-树脂体积比为40:1。

Extraction Process of Total Flavonoids from Spartina Alterniflora

ABSTRACT

Spartina alterniflora is a common herb, which comes from Atlantic coast of North America.

The purpose of this paper is to exploit the potential medicinal value of Spartina alterniflora, research on flavonids in Spartina alterniflora and prepare total flavonoids of Spartina alterniflora, so that the extraction rate of flavonids reaches an optimal level and the flavonids will be evaluated. This paper provide a reference for further exploit Spartina alterniflora.

The flavonoids were extracted by alcohol extraction, and the rutin standard solution was prepared with rutin and ethanol. To determine the maximum absorption wavelength and draw the rutin standard curve with the ultraviolet and visible spectrophotometer, Aluminum chloride, sodium acetate were used as color reagents, and ethanol was used as solvent. Single factor method was used to screen out the optimal concentration of ethanol , extraction temperature, the ratio of solid to liquid and extraction time. The prepared total flavonoids will be evaluated.

At the detection wavelength of 420nm, the equation of rutin standard curve is Y = 24.52X 0.033, where the R2=0.994. The optimal concentration of ethanol was 70%, the optimal extraction temperature is 60℃, the optimal ratio of solid to liquid is 1:30and the optimal extraction time was 90 minutes. After screening, AB-8 macroporous resin is adopted. Besides, the optimal adsorption time is 8 hours, desorption time is 6 hours, desorption liquid integral is 80 percent and desorption liquid-resin ratio is 40:1.
