利用正交法对苯胺降解菌MC-01降解条件优化的研究毕业论文

摘 要
苯胺是工业生产中的重要中间体,大量含苯胺及其衍生物的废水排入自然环境中,会对水域及周边土壤造成严重污染,危害人体健康。利用生物法处理苯胺废水,其经济高效而且无二次污染,但在生物处理过程中由于苯胺类废水独特的难降解性,引入高效菌种是大家一直关注的问题。本实验利用实验室自筛的苯胺降解菌MC-01,结合单因素实验优化降解条件,在此基础上,选用温度、pH和盐浓度三因素四水平进行正交实验设计,利用软件IBM SPSS statistics 19对苯胺降解条件组合进行优化分析,以sig值为参考指标,确定降解苯胺的最优条件组合。实验结果表明苯胺降解的最适条件组合是温度45℃,pH7.0,盐浓度为3%。在最适条件下验证,结果降解率为81.66%,大于正交实验中的所有处理组,进一步证实了该组合的降解率为最佳的,即为最适降解条件。

Study on the optimization of the degradation conditions of aniline degrading bacteria MC-01 by orthogonal method
ABSTRACT
Aniline is an important intermediate in industrial production, and a large amount of wastewater containing aniline and its derivatives is discharged into the natural environment, causing serious pollution to the soil and surrounding soil, and endangering human health. There are many methods to treat wastewater containing aniline, and the method of microbial degradation is an economical and effective method for treating aniline wastewater.. At present, the domestic and foreign scholars have carried out a lot of research on aniline degradation bacteria, and have been screened to many strains of highly effective bacteria. This experiment using laboratory screening self aniline degradation bacteria MC-01, combined with single factor experiments, the optimal degradation conditions, on the basis of this, the choice of temperature, pH and salt concentration on three factors and four levels orthogonal experiment design, using IBM SPSS statistics software of 19 aniline degradation conditions a combination of optimization analysis, as a reference index to determine the optimal conditions for the degradation of aniline by sig value. The experimental results showed that the optimum combination of aniline degradation was 45, pH7.0, and salt concentration of 3%. Under the optimum conditions, the degradation rate of the results was 81.66%, and the degradation rate of the composite was more than the orthogonal experiment, which was further confirmed that the degradation rate of the combination was the best, that was the optimum degradation conditions..
