摘 要
肿瘤坏死因子相关凋亡诱导配体属肿瘤坏死因子超家族成员(TNFSF10),表达于活化的T细胞等,有多个受体:TRAIL与DR4/DR5(胞质区含死亡结构域)结合可诱导细胞凋亡;TRAIL与DcR1/DcR2(缺乏胞质区或死亡结构域不完整)结合不能传递凋亡信号,并可抑制TRAIL诱导DR4/DR5阳性细胞凋亡的作用。
本研究实现了肿瘤坏死因子相关凋亡诱导配体进行基因在大肠杆菌中的克隆与表达。以肿瘤坏死因子相关凋亡诱导配体目的基因为模板,将其成功构建到pET22b载体中得到重组质粒pET22b-pelBtrail,并转化到E. coli BL21,得到重组菌E. coli BL21/pET22ba-pelBtrail,经菌落pcr验证后进行SDS-PAGE电泳。结果表明,重组菌E. coli BL21/pET22b-pelBtrail在20 kDa处呈现明显条带,与目的蛋白分子量一致,因此实现了肿瘤坏死因子相关凋亡诱导配体在大肠杆菌中的表达。
本研究实现了肿瘤坏死因子相关凋亡诱导配体实现在大肠杆菌中的高效可溶性表达,最大程度的获得可溶性trail蛋白,为肿瘤坏死因子诱导凋亡配体TRAIL在临床方面应用奠定了基础。

(Trail) clone of tumor necrosis factor-related apoptosis-inducing ligand in Escherichia coli
Abstract
Tumor necrosis factor related apoptosis inducing ligand belongs to tumor necrosis factor super family members (these two genes may), expressed on activated T cells, there are multiple receptors: combination of trail and DR4 / DR5 (cytoplasmic region containing the death domain) can induce cell apoptosis, trail and DcR1 / DcR2 (lack of cytoplasmic region or death domain is not complete) combination can not transmit apoptotic signals, and can inhibit the role of DR4 / DR5 positive cells apoptosis induced by trail.
In this study, the cloning of tumor necrosis factor related apoptosis inducing ligand gene in Escherichia coli and expression. To tumor necrosis factor related apoptosis induced ligand gene as a template, its successful construction to vector pET22ba recombinant plasmid pET22b-pelBtrail, and transformed to E. coli BL21. Recombinant E. coli BL21/pET22b-pelBtrail, by colony PCR verified by SDS-PAGE electrophoresis. The results show that, recombinant E. coli BL21/pET22b -pelBtrail showed a clear band at 29 kDa, which was consistent with the target protein molecular weight, so the expression of tumor necrosis factor related apoptosis inducing ligand in Escherichia coli was achieved.
In this study, the tumor necrosis factor related apoptosis inducing ligand in Escherichia coli, soluble expression was achieved, the greatest degree of soluble TRAIL protein, tumor necrosis factor induced apoptosis ligand (TRAIL) in clinical application to lay the foundation.
