糖蜜预处理工艺对Leuconostoc mesenteroides发酵联产葡聚糖-甘露醇的影响毕业论文

摘 要

葡聚糖和甘露醇被广泛应用于医药和食品等领域,其工业生产的改良也一直是研究热点。目前已经可以通过肠膜明串珠菌以蔗糖为底物发酵生产葡聚糖,以果糖为底物发酵生产甘露醇。但是成本依然巨大,极大地限制了产品的应用。糖蜜是工业生产蔗糖的副产物,含有大量的蔗糖、果糖、葡萄糖和生长因子,但由于还含有大量的杂质和有害离子,只能作废弃处理。若能以糖蜜代替蔗糖和果糖发酵生产葡聚糖和甘露醇,则能节省成本,解决制糖废料处理的问题。本实验采用了几种工业上常用的糖蜜预处理方法,分析了每种方法对糖蜜的影响,从而对比了哪种方法更适合肠膜明串珠菌发酵产葡聚糖和甘露醇。包括煮沸法[1]、硫酸法、活性炭法[2]、石灰法、磷酸法、亚铁氰化钾法[3]、PAM法和澄清法[4]

结果表明:硫酸法、PAM法和活性炭法均有效提高了葡聚糖的产物浓度,高于未处理的糖蜜。其中硫酸法处理过的糖蜜发酵效果最好,并且易于控制处理反应,适合应用于工业生产葡聚糖。煮沸法提高了甘露醇的产物浓度,操作简单,无添加试剂,适合应用于工业生产甘露醇。

The effects of molasses pretreatment on fermentation of glucan and mannitol by Leuconostoc enteroides

Abstract

Dextran and mannitol are widely used in medicine, food and other fields, which has also been a hotspot. Now we can produce dextran with the substrate of sucrose and mannitol with the substrate of fructose by Leuconostoc menteroides. But the cost is still great, which greatly limits its applications. Molasses is a byproduct from the production of sugar. And it contains a lot of sucrose, fructose, glucose and growth factors. But it can only be disposal because it also contains a lot of impurities and harmful ions. If we can produce dextran and mannitol using molasses instead of sucrose and fructose, it is able to save a lot of costs and also solve the problem of waste disposal. In this study, we used several commonly methods of molasses pretreatment in industry and analyzed the impact on molasses. Then we compared the methods, including boiling treatment, sulfuric acid treatment, activated carbon treatment, lime treatment, phosphate acid treatment, potassium ferrocyanide treatment, PAM treatment and clarification, to determine which method is the most suitable to produce dextran and mannitol by Leuconostoc menteroides.

The results showed that:Sulfuric acid treatment, PAM treatment and activated carbon treatment was effective to improve the product concentration of glucan, higher than the untreated molasses. Sulfuric acid treatment of molasses worked best and was easy to control treatment, so this method was suitable for industrial production. Boiling treatment improved the production of mannitol. Its operation was simple, without adding reagents. It was the most suitable method for industrial production.
